目的:研究大鼠脊髓持续性压迫后缩血管因素ET-3mRNA的改变,探讨痉、痿证方对大鼠脊髓持续性慢性损伤局部ET-3mRNA表达的影响。
方法:将雄性中年大鼠随机分为假手术组、轻压、中压及重压模型组和轻、中、重压的痉证方、痿证方治疗组。采用大鼠颈椎前路螺钉压迫颈脊髓,螺钉留置持续压迫30d以产生慢性损伤,用RT-PCR方法检测受压局部ET-3mRNA表达变化。
结果:正常脊髓组织中有ET-3mRNA表达,脊髓受压后ET-3mRNA表达明显增高,模型组与假手术组相比各组均P<0.01;ET-3mRNA表达增高程度与压迫程度有关,与轻压组相比,中、重压组差异具有显著性意义,P<0.01,重压与中压组相比,差异具有显著性意义,P<0.01;痉、痿证方干预后,增高的ET-3mRNA明显降低,与模型组相比,痉、痿证方组有统计学差异,P<0.01。
结论:痉、痿证中药方对增高的ET-3mRNA有明显的降低作用。 |

| Effects of Jing syndrome recipe(痉证方)and Wei syndrome recipe(痿证方)on ET-3mRNA expre ssion in injury local after constant com pression of spinal cord in rats |

ABSTRACT
Objective: To study the alteration of ET-3mRNA expr ession in rats'spinal cord after consta nt compressive injury,furthermore to exp lore the effects of Jing syndrome recipe (痉证方)and Wei syndrome recipe(痿证方)on ET-3 mRNA expression in injury local of spina l cord in rats.
Methods: The midlife male rats were randomly divi ded into four groups:sham-operated group ,model groups which included mild,modera te and severe compress groups,therapy gr oups of Jing syndrome recipe(痉证方)and Wei syndrome recipe(痿证方)which also included mild,moderate and severe compress groups .Screws were used to drill into spinal c o rd through anterior cervical vertebra bo dy.After operation the screws remained i n spinal cord for 30 days to create chro nic injury,and then RT-PCR was used to check the expression change s of ET-3mRNA in compressed location.
Results: There was expression of ET-3mRNA in normal spinal cord tissues.A fter spinal cord was compressed,the expr ession of ET-3mRNA increased significant ly( P <0.01).The increase degrees of ET-3 mRNA expressions were related to the deg ree of compression,and the expressions w ere different in the mild,moderate and s evere compress groups( P <0.01).After Jin g syndrome recipe(痉证方)and Wei syndrome recipe(痿证方)were used,the increased ET-3m RNA expression decreased significantly( P <0.01).
Conclusion: Jing syndrome recipe(痉证方)and Wei syndrome recipe(痿证方)are effective to decrease ET -3mRNA expression in injuried spinal cord after constant compression. |
