目的:探讨黏附因子Ncadherin在骨髓间质细胞诱导分化中的表达及其转化机制。
方法:体外分离,扩增Wistar大鼠骨髓间质细胞,按一定浓度种植于24孔板内,并加入TGFβ诱导剂,于不同时间点取材固定。用免疫组化方法动态观察Ncadherin在骨髓间质细胞诱导分化中表达时相。同时加入抗Ncadherin抗体干扰细胞聚集。
结果:Ncadherin主要表达于加入诱导剂后24h,之后呈下调控趋势,并呈现时间依赖性。加入抗体后可明显干扰细胞聚集,当浓度为300μl/孔时,未见细胞小结形成。
结论:Ncadherin介导了细胞与细胞之间的相互作用,在体外间质细胞转化中发挥重要作用。 |

| Expression of N-cadherin in mesenchymal stem cells by inducing differentiation of TGF-β |

ABSTRACT
Objective:To investigate the expression of N-cadherin by inducing differentiation of TGF-β 1.
Methods:The mesenchymal stem cells of Wistar Rats were harvested in vitro and implanted in 24-hole plates in a monolayer culture as well as fixed at different timepoint after adding inducer TGF-β1,immunohistochemistry was applied to observe the time-phase of expression of N-cadherin during cell transformation,and with its anti-body to disturb aggregation of cells.Result:N-cadherin was expressed only in 24h after adding TGF-β1,and then down-regulation and time-dependant phase were be showed,its antibody could inbibit the cellular aggregation completely.The optimal dose was 300 ul/hole.
Conclusion:N-cadherin mediates interaction among cells,it is an important role playing in the differentiation of cells. |
