健脾益气法对严重创伤软组织损伤创面组织中碱性成纤维细胞生长因子和表皮生长因子的影响

目的:采用眼眶静脉放血结合刀割法制备严重创伤的动物模型,观察创面修复过程中创面组织中碱性成纤维细胞生长因子(bFGF)以及表皮生长因子(EGF)表达,研究健脾益气法促进严重创伤大鼠软组织修复可能的作用机制。

方法:40只SD健康成年大鼠,采用眼眶静脉放血结合刀割法制备严重创伤软组织损伤模型,然后将存活大鼠(33只)随机分为健脾益气组、活血化瘀组、模型组。各组大鼠均在第3、7、14天取部分创面及少许周围的正常皮肤,采用免疫组织化学染色后观察创面肉芽组织中bFGF和EGF表达。

结果:造模后3、7、14 d,健脾益气组、活血化瘀组大鼠创面组织中bFGF和EGF表达均较模型组大鼠创面组织中的含量明显增强。与活血化瘀组相比较,造模后3、7 d,健脾益气组大鼠创面组织中bFGF和EGF含量较高(P<0.05);但造模后14 d,两组大鼠创面组织中bFGF和EGF的含量无明显差异(P>0.05).

结论:健脾益气法具有增强严重创伤软组织损伤创面组织中bFGF和EGF表达的作用。

Effect of reinforcing Qi strength spleen(健脾益气) in the expression of bFGF and EGF in treating serious soft tissue injury
Abstract:

Objective: Using the method of bleeding from the orbital vein and lancing to make the animal model of trauma,and to observe the influence of reinforcing Qi strength spleen(健脾益气)in the expression of bFGF and EGF in the reparative process of raw surface,in order to explore the possible mechanism of reinforcing Qi strength spleen(健脾益气)in promoting the rehabilitation of soft tissue.

Methods: Forty healthily adult SD rats were made to be traumatic model using the method of bleeding from the orbital vein and lancing. After operation,there were 33 rats survival,which were divided into the reinforcing Qi strength spleen(健脾益气)group,the activating blood circulation to dissipate blood stasis(活血化瘀) group and the model group randomly. The raw surface and ambient normal skin were taken at the 3rd,7th and 14th days after operation to detect the expression of bFGF and EGF by immunohistochemical method.

Results: At the 3rd,7th and 14th days after operation,the expression of bFGF and EGF in the tissue of raw surface of the reinforcing Qi strength spleen(健脾益气)group and the activating blood circulation to dissipate blood stasis(活血化瘀) group was obviously higher than that of the model group(P<0.05). Compared with the activating blood circulation to dissipate blood stasis(活血化瘀) group,the expression of bFGF and EGF in the tissue of raw surface of the reinforcing Qi strength spleen(健脾益气)group was higher(P<0.05) in the 3rd and 7th day after operation. But in the 14th after operation,there was no significantly difference between reinforcing Qi strength spleen(健脾益气)group and activating blood circulation to dissipate blood stasis(活血化瘀) group.

Conclusion: The method of reinforcing Qi strength spleen(健脾益气)can efficiently promote the expression of bFGF and EGF in raw surface of serious soft tissue injury.
