恒古骨伤愈合剂对激素性坏死股骨头内VEGF基因表达的影响

目的:研究中药恒古骨伤愈合剂对兔坏死股骨头内VEGF基因表达的影响。

方法:成年新西兰大白兔分为4组:A组(空白对照组),B组(正常兔服用恒古骨伤愈合剂),C组(造模后不喂恒古骨伤愈合剂),D组(造模后喂恒古骨伤愈合剂),每组24只。以大肠杆菌内毒素(LPS)(10μg/kg体重)间隔24h静注2次加甲基强的松龙(MPS)肌注3次制作兔股骨头坏死模型后,B组和D组用恒古骨伤愈合剂,A组和C组用等量生理盐水,按计算出的药量灌胃,1次/2d。于喂药至第4、8、12周时取股骨头行免疫组化、原位杂交并提取总RNA行实时荧光定量PCR,动态观察兔股骨头内VEGF基因表达的变化特点。

结果:喂药至第4、8周时,比较D组与C组股骨头内VEGF基因表达量的差别无统计学意义。喂药至第12周时,D组与C组股骨头内VEGF基因表达量的差别具有统计学意义(P=0.042<0.05)。B组与A组比较差别无统计学意义。

结论:LPS加MPS制作的兔股骨头坏死模型中股骨头内VEGF基因表达受抑制。中药恒古骨伤愈合剂可促进坏死股骨头内源性VEGF基因的表达,以用药时间长者效果明显。恒古骨伤愈合剂对正常兔股骨头内VEGF基因的表达无影响。

Effects of the VEGF gene expressions of Osteoking in the treatment of femoral head necrosis
Abstract:

Objective:To study the effects of vascular endothelial growth factor(VEGF) gene expressions of Osteoking in the treatment of femoral head necrosis(FHN).

Methods:Rabbits were divided into 4 groups randomly:group A(blank control group),group B(normal rabbit fed with Osteoking),group C(model group without feeding Osteoking),group D(model group fed with Osteoking).There were 24 rabbits in each group.Rabbit model of FHN was established with the method of twice intravenous injection with LPS(10 μg/kg body weight,at a time interval of 24 hours)and 3 times intramuscular injection with Methylpredniso lone(MPS)(20 mg/kg body weight,at a time interval of 24 hours).Once every 2 days,the stomach in group B and D were irrigated with Osteoking,while A and C with normal saline.After the treatment with Osteoking,the changes in VEGF genes expressions were compared with each group.At the 4th,8th and 12th week,8 rabbits were put to death in each group.Then we took out both femoral heads,make immunohistochemistry,in situ hybridization and extract total RNA for real-time Q-PCR.The dynamic changes of the VEGF gene expressions were successfully detected inside the femoral head of rebbits.

Results:There were no significant difference in expressions of the VEGF gene between model group without feeding Osteoking and model group fed with Osteoking at the 4th week and 8th week(P>0.05),but there were obviously difference at the 12th week(P=0.042).The expressions had no difference in blank control group and normal rabbit fed with Osteoking group at different time.

Conclusion:The VEGF gene expressions are restrained in LPS and MPS model.Osteoking can stimulate the endogenous VEGF gene expressions of FHN;and the longer the time it has heen used,the better the effect is obvious.Osteoking has no effect to the femoral head VEGF gene expressions for normal rabbit.
