目的:研究间歇性压力培养环境对兔骨髓基质干细胞(MSC)增殖的影响,探讨假体置换术后活动所需的最适宜压力。
方法:以新西兰兔的骨髓基质干细胞为实验材料,通过噻唑盐(MTT)比色试验,观察间歇性压力对骨髓基质干细胞增殖的影响。
结果:MSC在60、100kPa间歇性压力培养环境下MTT反应的OD值小于对照组,压力值越大,OD值越小(P<0.05)。而20kPa间歇性压力情况可显著促进MSC增殖,并随时间增加而增强。在实验第7天时,实验组OD值高于对照组(P<0.05),差异有显著性意义。
结论:关节置换术后早期下地活动产生的较大应力,会明显抑制骨髓腔内MSC的增殖,不利于骨的重建愈合,临床应当避免。然而,较小的间歇性压力对MSC增殖有明显的促进作用,提示关节置换患者可在较小压力范围(<20kPa)内进行早期关节被动活动训练,有利于关节术后骨的重建愈合。 |

Osteoblast proliferation of rabbits bone marrow stromal cells cultured under intermittent stress |

Abstract:
Objective: To study the effect of bone marrow stromal cells(MSC) on different intermittent stress and explore the optimum pressure for the activity after prosthesis replacement.
Methods: Using MTT testing,osteoblast of rabbits bone marrow stromal cells cultured under controlled intermittent stress environment,which proliferation was measured.
Results:Under 60,100 kPa intermittent stress,optical density(OD) was similar with that under continual stress.However,under 20 kPa intermittent stress,the OD was higher than control group(P<0.05).The longer period of observation was,the higher OD was.
Conclusion: Early out-of-bed activity after total hip replacement operation,may bring about major stress on MSC in medullary cavity and is harmful of patient reestablish.But minor stress can accelerate MSC proliferation.This indicates that after total hip replacement operation,early on-of-bed activity may bring about intermittent stress on MSC in medullary cavity and is beneficial of patient rehabition. |
