目的:研究铸骨胶囊对去势大鼠骨质疏松症的治疗作用。

方法:选用雌性6月龄SD大鼠60只,随机分为Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ、Ⅵ组(分别为假手术组,模型组,模型+骨疏康颗粒组,模型+铸骨胶囊高、中、低剂量组),Ⅰ组在双侧卵巢位置切除与卵巢同等大小的脂肪块,其他各组(Ⅱ~Ⅵ)切除双侧卵巢造成绝经后骨质疏松症动物模型。Ⅰ、Ⅱ组按1ml/100g体重灌胃(1.33%的β-环糊精水溶液),Ⅲ组按2.7g/kg体重灌胃(骨疏康颗粒水溶液),Ⅳ、Ⅴ、Ⅵ组分别按400、200、100mg/kg体重灌胃(铸骨胶囊水溶液)。给药3个月后,检测各组动物的左侧股骨及第2腰椎骨密度(bonemineraldensity,BMD),股骨生物力学强度(用三点弯曲实验检测),对胫骨切片进行骨组织形态计量学分析,采用放免法测血清中雌二醇(estradiol,E2)、降钙素(calcitonin,CT)、骨钙素(bonegalprotein,BGP)含量。

结果:切除雌性大鼠双侧卵巢3个月后,腰椎及股骨BMD明显降低,与Ⅰ组比差异有显著性统计意义(P<0.05),提示绝经后骨质疏松症动物模型复制成功。铸骨胶囊各组(Ⅳ、Ⅴ、Ⅵ)与Ⅱ组比较,Ⅳ组腰椎和股骨BMD增加,差异有显著性统计意义(P<0.05,P<0.01)。Ⅳ组股骨生物力学强度各项指标及胫骨骨组织形态计量参数明显改善,差异有显著性统计意义(P<0.05);CT、BGP含量升高,差异有显著性统计意义(P<0.001,P<0.01)。Ⅳ组与Ⅲ组比较,最大桡度增加,差异有显著性统计意义(P<0.05);骨小梁形成表面(trabeculaformationsurface,TFS)百分比增加,差异有显著性统计意义(P<0.01);CT、BGP含量升高,差异有显著性统计意义(P<0.05,P<0.01)。

结论:铸骨胶囊可以增加切除双侧卵巢大鼠的BMD,提高骨的力学性能,降低骨折的发生率,对去势大鼠有一定的抑制骨丢失作用。

Study the effects of Zhugu(铸骨) capsule on post-menopausal osteoporosis in ovariectormized rats
Abstract:

Objective:To observe the effects of Zhugu(铸骨) capsule on osteoporotic rats caused by ovariectomizy.

Methods:Sixty female SD rats at the age of six-months were randomly divided into six groups (10 rats every group):Group Ⅰ(sham operation group),Ⅱ(ovariectomizy group),Ⅲ(ovariectomizy+Gushukang granule group),Ⅳ(ovariectomizy+high-dose Zhugu capsule group)、Ⅴ(ovariectomizy+middle-dose Zhugu capsule group),Ⅵ(ovariectomizy+low-dose Zhugu capsule group).The axungia pieces of the ovaries position were excised in the group Ⅰ.The ovaries were excised in the other groups to make animal model of post-menopausal osteoporosis.Group Ⅰ,Ⅱ were fed with 1.33% β-cyclodxtrin at the dose of 1 ml/100 g,group Ⅲ was fed with Gushukang(骨疏康) granule at the dose of 2.7 g/kg,group Ⅳ,Ⅴ,Ⅵ were fed with Zhugu(铸骨) capsule at the dose of 400,200,100 mg/kg respectively.Bone mineral density(BMD) of the left femur and the second lumbar vertera,biomechanical properties of femur by three-point bending test,bone histomorphometric analysis of tibia,serum contents of estradiol (E2),calcitonin (CT) and bone gal protein (BGP) by the radioimmunoassay (RIA) were measured respectively at the end of the 3rd month.

Results:The BMD of the left femur and the second lumbar vertebra were decreased after ovariectomizy rats (P<0.05),which demonstrated the an-imal model of post-menopausal osteoporosis was established successfully. As compared with the group Ⅱ, theBMD of lumbar and femur in group Ⅳ were increased (P<0.05,P<0.01);the biomechanical properties of femur were increased (P<0.05);the histomorphometric analysis of the tibia had significantly improved (P<0.05);the serum contents of CT and BGP were increased (P<0.001,P<0.01).As compared with group Ⅲ,the maximum deflection in group Ⅳ was increased (P<0.05);the pet of trabecula formation surface (TFS) in group Ⅳ was increased(P<0.01);the serum contents of CT and BGP in group Ⅳ were increased (P<0.05,P<0.01).

Conclusion:Zhugu capsule can increase BMD of ovariectomized rats,improve the biomechanical properties of ovariectomized rats and reduce the incidence rate of fracture,which have inhibition for bone loss of ovariectomized rats.
