缺氧对成人成骨细胞增殖及分化的影响

目的:在缺氧条件下体外培养成人成骨细胞,探讨缺氧对成骨细胞增殖及分化的影响。

方法:将手术中取得的成人髂骨骨块,使用酶消化法进行培养,当细胞传至第2代时,建立成骨细胞缺氧模型,用MTT法观察各组成骨细胞增殖活力的变化;用骨钙素放射免疫法分别检测各组骨钙素含量;用激光共聚焦显微镜对两组培养前3d细胞血管内皮生长因子(VEGF)光密度值作定量分析。

结果:正常组细胞增殖活力较缺氧组强;缺氧组细胞VEGF的光密度值及骨钙素含量均高于正常组。

结论:缺氧抑制成骨细胞增殖,缺氧(1~3d)诱导成人成骨细胞VEGF的高表达并促进成骨细胞的分化。

Effects of hypoxia on osteoblast proliferation and differentiation
Abstract:

Objective:To explore the effects of hypoxia on osteoblasts proliferation and differentiation.

Methods:Obtained osteoblasts from the adults iliac bone during operation were cultured by the method of enzyme digestion in vitro.Hypoxic model of osteoblast were established when cell subculturing into the second generation.The cell proliferation and osteocalcin(OCN) content were detected by MTT and radioimmunoassy,respectively.The VEGF optical density(O.D.) value of cells(which were cultured for the first three days) were rationally analyzed by laser confocal microscope.

Results:The cell proliferation was inhibited in the hypoxic group when the cells were cultured for the first three days.The O.D.value of VEGF and OCN content of the hypoxic group were more than that of the normal group.

Conclusion:Hypoxia can inhibit the osteoblasts proliferation.Hypoxia(1-3 days) can induce the high expression of VEGF and advance the osteoblasts differentiation.
