骨髓基质细胞与可降解明胶-聚羟基丁酸酯膜复合培养的生物合成功能分析

目的:从生物合成功能角度,探讨骨髓基质细胞(BMSCs)与可降解明胶-聚羟基丁酸酯(G-PHB)膜复合培养的生物相容性。

方法:以BMSCs分别与G-PHB、聚羟基丁酸酯(PHB)、明胶(G)膜材料复合培养,并设立空白对照组,各组通过3H-TdR法检测DNA合成;考马斯亮蓝法检测蛋白质合成;对硝基苯磷酸盐法(PNP)测定碱性磷酸酶(ALP)水平;放免法测定骨钙素(OCN)表达;流式细胞术测定纤维连接蛋白(FN)表达。

结果:G-PHB组DNA合成优于PHB组和空白对照组(P<0.05)。总蛋白合成与各对照组无显著性差异(P>0.05),但成骨特异性标志ALP、OCN以及细胞黏附指标FN均明显优于其他各组(P<0.05)。

结论:G-PHB材料对于种子细胞BMSCs黏附、增殖与成骨定向分化有明显的促进作用,是一种有效上调细胞生物合成功能的新型可降解材料。

Analysis of biosynthetic function on mixed culture of BMSCs and absorbable G-PHB membrane
Abstract:

Objective:To investigate biological compatibility on mix culture of BMSCs and absorbable G-PHB membrane basing on standpoint of biosynthetic function.

Methods:BMSCs were cultured with G-PHB,PHB and Glutin(G) membrane respectively to establish the blank control group.In the groups,the DNA and protein synthesis,Akaline phosphatase(ALP) level,Osteocalcin(OCN) and fibronection(FN) expression of BMSCs were respectively detected by 3H-TdR,Coomassie brilliant blue staining,paranitro-pheneye phosphate(PNP),radioimmunoassay(RIA),flow cytometry(FCM).

Results:The DNA synthesis in the G-PHB group was higher than that of the PHB and the blank control group(P<0.05).There was no significant difference in total protein synthesis were observed G-PHB group and other groups(P>0.05).The ALP level,OCN and FN expression in G-PHB group were better than those of groups(P<0.05).

Conclusion:The G-PHB has obviously auxo-action for adhesiveness and proliferation of BMSCs,and osteogenesis oriented differentiation.It is a new pattern and absorbable material which can effective cause up-regulation of biosynthesis function of cell.
