目的:探讨活血化瘀汤对骨折早期缺氧诱导因子1α(HIF-1α)mRNA和血管内皮生长因子(VEGF)表达的影响及骨折早期缺氧反应的机制。
方法:选用雄性SD大鼠72只,造成左胫骨中段闭合折骨标准的骨折模型,术后采用髓内针固定。随机分成活血化瘀汤组(麻醉苏醒灌服活血化瘀汤,n=36)和生理盐水组(麻醉苏醒灌服等量生理盐水,n=36),分别于灌胃后1、2、3、5、7、14d处死大鼠,取包括骨折端在内骨折上下各0.5cm的骨质。应用TRPCR技术,动态观察HIF-1αmRNA及VEGF表达的变化。
结果:在骨折后2d,骨折端HIF-1αmRNA表达达到高峰;活血化瘀汤组用药后1d及2d,骨折端HIF-1αmRNA表达比生理盐水组显著性降低(P<0.01);用药后3、5、7、14dVEGF的表达活血化瘀汤组比生理盐水组显著性升高(P<0.01);正常对照组对应组织则不能检测到两者的表达。
结论:左胫骨中段闭合骨折引起HIF-1αmRNA及VEGF基因转录和表达发生改变。活血化瘀汤能调控HIF-1α和VEGF的表达。 |

Experimental research on the influence of Huoxue Huayu decoction(活血化瘀汤) to HIF-1α mRNA and VEGF expression during early fracture healing |

Abstract:
Objective:To explore the expression of hypoxic inductive factor-1α mRNA (HIF-1α mRNA) and vascular endothelial growth factor (VEGF) during early fracture healing of rats with Huoxue Huayu decoction(HXHYD,活血化瘀汤).
Methods:Sprague-Dawley rats(n=72)were caused standardized closed left tibial fracture under isoflurane-anesthetize with a modified technique.36 rats were treated with HXHYD after fracture.The other 36 rats without medicine served as controls.Fractured sides were determined with HIF-1α mRNA and VEGF expression on 1,2,3,5,7 and 14 d.Three normal rats without any medicine served as normal controls.
Results:The expression of HIF-1α mRNA was increased significantly on the fracture sides 2 days after fracture but decreased significantly after treatment with HXHYD on 1,2 d(P<0.01).In the same time,the expression of VEGF was increased significantly after treatment by HXHYD on 3,5,7 and 14 d(P<0.05).The expression of HIF-1α mRNA and VEGF were not detected in normal control group.
Conclusion:This study has confirmed that fracture leads to microcirculation failure in both fracture sides and related skeletal muscle.HXHYD can improve blood nutrition on fractured sides during early fracture healing via the balance of HIF-1α mRNA and VEGF expression. |
