补肾活血中药对体外培养成骨细胞增殖的影响 |

目的:探讨补肾活血中药对胎鼠颅盖骨来源成骨细胞增殖的影响,以进一步了解补肾活血中药改善骨代谢的作用机制。
方法:取二次酶消化法培养的胎鼠颅骨第3代成骨细胞,随机分为5组:细胞对照组、空白血清组、含药血清组、原药组、阳性对照药(αD3)组,分别加入普通细胞培养液、含空白血清及不同浓度补肾活血中药含药血清、补肾活血中药原药液、阳性对照药(αD3)的条件培养液,在培养第3、6天用MTT比色法观察补肾活血中药含药血清及原药液对成骨细胞增殖率的影响。
结果:补肾活血中药含药血清组成骨细胞增殖率较空白血清组、培养液组、αD3组显著增强,且存在剂量依赖关系,随培养期延长,促进作用持续存在。补肾活血中药原药液在一般浓度对成骨细胞增殖无显著影响,而过高浓度对成骨细胞增殖有抑制作用。
结论:补肾活血中药含药血清能够直接作用于体外培养的成骨细胞。 |

Effects of tonify-kidney and activate-blood Chinese medicine on proliferation of rat’s osteoblasts in vitro |

Abstract:
Objective:To investigate the effects of tonify-kidney and activate-blood Chinese medicine on rat osteoblasts in vitro,and to understand the mechanism how the Chinese medicine promotes bone formation.
Methods:The osteoblasts were isolated from newbone rat’s calvaria by sequential enzyme digestion,and the cells of the third generation were divided into five groups randomly and treated with different culture medium.After 3 and 6 days incubation the effects of tonify-kidney and activate-blood Chinese medicine on proliferation of the osteoblasts were observed with MTT assay.
Results:The tonify-kidney and activate-blood Chinese medicine combined with the serum could stimulate the proliferation of the osteoblasts in dose-dependent manner.With the time of the incubation,the stimulation persistly exists.High dose of the tonify-kidney and activate-blood Chinese medicine inhibits the proliferation of the osteoblasts.
Conclusion:The tonify-kidney and activate-blood Chinese medicine can improve the proliferation of the osteoblasts. |
