rhBMP-2对兔骨髓基质细胞VEGF表达及成骨潜能的影响

目的:观察经不同剂量重组人骨形态发生蛋白2(recombinant human bone morphogenetic protein2,rhBMP-2)刺激后的兔骨髓基质细胞(marrowstromalcell,MSC),其血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)表达及成骨潜能变化的情况。

方法:取兔双侧股骨骨髓基质细胞体外培养,分别以不同剂量rhBMP-2刺激细胞,并设立空白对照组。培养5d后,进行细胞形态(HE染色)、增殖情况(细胞记数法与MTT法)、碱性磷酸酶(组化染色与活性测定)、钙化结节(图像分析)、VEGF阳性细胞率(免疫组化染色)等项目的检测。

结果:不同剂量组间在碱性磷酸酶活性、矿化面积百分率、VEGF阳性细胞率三项观测指标上差异显著,rhBMP-2剂量为100ng/ml左右时效果最佳;在细胞记数与MTT检测上各组差别不显著。

结论:应用rhBMP-2在促进基质细胞成骨潜能的同时,还可促进VEGF的表达,其应用剂量与效果之间存在明显相关关系,应用剂量为100ng/ml左右时,其促进VEGF表达及成骨潜能作用同时达到最佳效果,超过此应用剂量,两者则有下降趋势。

Effects of different dose of rhBMP-2 on VEGF expression and osteogenic potential of rabbits bone marrow stromal cells
Abstract:

Objective:To investigate the effects of different dose of ecombinant human bone morphogenetic protein-2(rhBMP-2r) on vascular endothelial growth factor (VEGF) expression and osteogenic potential of rabbit bone marrow stromal cells.

Methods:The marrow stromal cells (MSC) of the rabbit’s femur were procured and cultured,the samples were treated with different dose of rhBMP-2,and without treation as control.Five days later,the effects were observed with histopathological analysis,cells proliferation,alkaline phosphatase activities detection,the computer analysis of bone clusters,detection of ratio VEGF-positive cells (immunohistochemistry).

Results:There was statistically significant difference between different doses of groups in alkaline phosphatase activities detection,the computer analysis of bone clusters and detection of ratio VEGF-positive cells,the group treated with 100 ng/ml rhBMP-2 had the best effects in these items.We found no statistically difference between different dose groups in other items.

Conclusion:rhBMP-2 is a wonderful osteoinductive reagent,when it is used to stimulate marrow stromal cells,both VEGF expression and osteogenic potential of the cells advance,and there are obvious relative relationship between doses and effects.The effects also indicate that the ability of rhBMP-2 enhancing VEGF expression and osteogenic potential reached to its highest point,when applying dose of rhBMP-2 is 100 ng/ml,when exceeding the dose,it has a downtrend.There are no obervious effects of rhBMP-2 on marrow stromal cells’proliferation.
