坐骨神经创伤性华勒氏变性中雪旺细胞凋亡初步研究 |

目的:探讨成年大鼠坐骨神经创伤性华勒氏变性中雪旺细胞凋亡的存在及其时相性变化规律。
方法:利用成年雌性SD大鼠坐骨神经横断复制华勒氏变性模型。将74只SD大鼠随机分为12组。1个正常组(8只)、11个实验组(66只)。每只实验组大鼠双后肢再随机分为试验肢(构成试验组)和对照肢(构成对照组)。分别于术后1h,6h,12h,24h,2d,3d,4d,8d,14d,21d,30d共11个时相点,切取坐骨神经中下段标本。检测各组神经纤维形态学改变,雪旺细胞S100蛋白和凋亡基因相关产物Bcl2、Fas、Bax表达水平变化,并用TUNEL法检测雪旺细胞凋亡指数。
结果:①试验组雪旺细胞数量术后24h开始明显减少,术后8d才开始重新增多。②S100蛋白表达对照组变化无统计意义(P>:005),试验组则在术后1h,21d两个时间点变化有统计意义(P<:005)。③雪旺细胞凋亡抑制基因产物Bcl2表达,正常组双侧肢变化无差异(P>:005),而对照组术后6h明显升高,术后24h、4d、21d轻度增高(P<:001),试验组术后6h,24h,8d,14d,30d进行性升高(P<:001)。④雪旺细胞凋亡促进基因产物Bax表达,正常组双侧肢比较无差异(P>:005),而对照组术后6h,3~21d持续高表达(P<:001),试验组14d开始。 |

Preliminary investigation on Schwann cell apoptosis in Wallerian-degenerated sciatic nerve of rat |

Abstract:
Objective:To investigate existence and regularity of changes of Schwann cell apoptosis in Wallerian degenerated sciatic nerve of rat,and evaluate its time-related feature.
Methods:45 SD rats were divided randomly into one normal group (8 rats) and eleven experimental groups(66 rats).Both hind-legs of each rat in experimental groups were randomly divided into test leg(with sciatic nerve transection) and control leg(uninjured nerve).All test legs constituted a test group and all control legs constituted a control group.After operation,all rats were respectively sacrificed at 1 h,6 h,12 h,2 d,3 d,4 d,8 d,14 d,21 d and 30 d.The specimens of mid distal sciatic nerve,especially the morphological changes of the nerve,the different expression levels of S-100 protein and apoptosis related proteins such as Bcl-2,Bax,and Fas in Schwann cell were be analyzed.The TUNEL method was used to detect apoptotic index of Schwann cell rate.
Results:①The test group showed Wallerian degeneration.The number of Schwann cell began to decrease at 24 h,surprisingly be rare at day 3 and 4,then began to increase from day 8 and formed Bungner belt after 14 days.②Schwann cell generally expressed S-100 at low level in all groups.The control group was not significantly different from the normal group;There was statistical significance in the test group at 1 h and day 21.③Bcl-2 positive rates of the control group and the test group apparently elevated and were significantly different from the normal group.④The control group and the test group expressed Bax at high level and were significantly different from the normal group.⑤Fas positive rate of the control group was slightly elevated,but without statistical significance compared with the normal group;Fas positive rate of the test group continuously elevated in fluctuant way,with highly statistical significance compared with the normal group.⑥There was rarely existed Schwann cell apoptosis in the normal group,and no statistical significance of the left legs compared with the right legs;The test group showed lots of apoptotic nuclei at 6 h,2 d,4 d and 21 d.There was highly statistical significance of cell apoptosis compared with the normal group.
Conclusion:Schwann cells are existed in Wallerian degeneration of rat sciatic nerve after transection,with time-related feature. |
