人类骨肉瘤中P53、Bcl-2、Bax蛋白的表达和临床特征

目的:研究P53、M-2和Bax蛋白在骨肉瘤中的表达情况,探讨P53、Bcl-2和Bax基因在骨肉瘤发生和发展中的作用,研究三者在骨肉瘤中的表达与骨肉瘤组织学分级之间的关系。

方法:采用SP免疫组化方法观察42例骨肉瘤中P53蛋白、BcLZ蛋白和Bax蛋白的表达情况。

结果:1.在骨肉瘤中P53蛋白、el-2蛋白和ux蛋白的阳性率分别为61.9%、66.7%、61.9%。2.骨肉瘤I~皿级中P53蛋白阳性率分别为41.7%、53.3%石5.7%,在1级与Ill级之间存在显著性差异(P<0刀5)。3.骨肉瘤I一皿级中Bcl-二蛋白阳性率分别为41.7%、62.5%、92.9%。在11级与皿级之间存在显著性差异(P<0.05),在1级与Ill级之间存在显著性差异(P<0.01)。Bax蛋白阳性率在各级间无显著性差异(P>0.05)。4.在骨肉瘤P53蛋白表达阴性组中el-2蛋白的阳性率显著低于P53蛋白表达阳性组炉<0.01)。5.在骨肉瘤Bcl-2蛋白表达阴性组中Bax蛋白的阳性率显著低于el-2蛋白表达阳性组(P<0.05)。

结论:利用SP免疫组化法检测骨肉瘤中P53、el-2和Bax蛋白的表达具有较高的敏感性及临床实用价值。

Expression and clinical significance of P53.Bcl-2 and Bax protein in human osteosarcoma
Abstract:

Objective:study the expression of P53, Bcl-2 and Bax protein in osteosarcoma, and dis-cuss the function of P53,Bcl-2 and Bax gene in the occurrence and development of human osteosarcomas.to study the relation between the expression of the three proteins and the pathological classification in osteosarco-mas.

Methods:42 cases of osteosarcoma have been examined with the method of S-P immunohistochemistry.

Results:1.The expression rate of P53, Bcl-2 and Bax protein in osteosarcoma was respectively 61.9%, 66.7% and 61.9% .2.The expression rate of P53 protein in osteosarcoma grade Ⅲ was higher than that in osteosarcoma grade I (P<0.05).3.The expression rate of Bcl-2 protein in osteosarcoma grade Ⅲ was higher than that in osteosarcoma grade I (P<0.01) ,but the expression rate of Bax protein had no significant dif-ference(P>0.05).4.The expression rate of Bcl-2 protein in cases with negative P53 protein expression was lower than that in cases with positive P53 protein expression(P<0.01).5.The expression rate of Bax protein in cases with negative Bcl-2 protein expression was lower than that in cases with positive Bcl-2 protein expres-sion(P<0.05).

Conclusion: Expression of P53,Bcl-2 protein is related to the pathological classification and can reflect the malignant degree of osteosarcoma. S-P imrnunohistochernistry method has a higher sensitivity and clinical value.
