目的:探讨外源性降钙素基因相关肽(CGRP)对大鼠颅盖骨来源成骨细胞的影响,以进一步了解CGRP促进成骨的作用机制。

方法:利用二次酶消化法培养的成骨细胞,加入含不同浓度CGRP的条件培养液,24小时后通过MTT比色,3H-TdR掺入以及细胞内碱性磷酸酶(ALP)含量的测定来观察CGRP对成骨细胞增殖和分化的影响。

结果:CGRP可以明显促进成骨细胞增殖,而且作用呈剂量依赖性。CGRP对细胞内ALP含量无明显影响。

结论:CGRP能够直接作用于成骨细胞并通过促进其增殖而有利于骨形成。

The effects of calcitonin gene-relate peptide on proliferation and differentiation of rat osteoblasts
Abstract:

Objective:To explicate the effects of exogenous calcitonin gene-related peptide(CGRP)on rat osteoblasts in vitro,and to investigate the mechanism by which CGRP promotes bone formation.

Methods:The osteoblasts were isolated from newborn rat calvaria by sequential enzyme digestion,and the secondary cells were treated with different concentrations of CGRP respectively.After 24 hours of incubation,the effects of CGRP on proliferation and differentiation of osteoblasts were observed with MTT assay, 3H-TdR incorporation and the determination of intracellular ALPase activity.

Results:CGRP could stimulate the proliferation of osteoblasts in dose-dependent manner,but was unable to affect the intracellular ALPase activity of osteoblasts.

Conclusion:CGRP can act directly on osteoblasts and increase bone formation by stimulating the proliferation of osteoblasts,but it does not promote the differentiation of osteoblasts.
