目的:从分子水平认识转化生长因子β1(TGFβ1)在骨折愈合过程中的作用。

方法:采用地高辛(DIG)标记寡核苷酸探针的原位杂交方法,光镜观察成年新西兰白兔桡骨中段骨折愈合过程中,TGFβ1、mRNA在骨痂区各组织细胞中的表达变化情况。

结果:在骨折愈合早期,可见少量TGFβ1表达阳性的单核细胞、间充质细胞和血管内皮细胞。在膜内成骨期,TGFβ1在成骨细胞内有显着表达,尤其是多角形丰满成骨细胞。在成软骨期,较成熟软骨细胞内有较高表达。在软骨内成骨期,成骨细胞内有高表达,而在肥大的软骨细胞内几乎无表达。

结论:TGFβ1在整个骨折愈合过程中的每个阶段都具有重要作用,影响着细胞分化、增殖与成熟以及基质的形成。

Expression of transforming growth factor β1 during fracture healing
Abstract:

Objective:To investigate the role of transforming growth factor β1(TGF β1)in fracture healing on the molecular level.

Methods:The local expressions of TGF β1 in callus during the process of healing of adult New Zealand white rabbits were investigated through optical microscope,using in situ hybridization of DIG labeled oligonucleotide probe.

Results:At the early stage of fracture healing,TGF β1、mRNA were presented in a few monocytes,mesenchymal cells,and endothelial cells.At the stage of intramembranous bone formation,TGF β1 expressed notably in the osteoblasts,especially in the multiangle plentiful osteoblasts.At the stage of chondrogenesis,nearly mature chondrocytes were strongly positive.While at the stage of endochondral ossification,the hypertrophic chondrocytes were almost negative.TGF β1 expressed strongly in osteoblast at this stage.

Conclusion:TGF β1 acted as an important role in every stage of fracture healing by affecting the differentiation,proliferation,maturation of cells and the formation of matrix.
