中文摘要:
目的: 探讨甲状旁腺素(PTH)对人骨关节炎软骨细胞终末期分化的抑制作用.
方法: 分离和培养人骨关节炎软骨细胞,将其进行分组(PTH处理组和对照组),分别经PTH和盐水处理细胞,倒置显微镜观察细胞形态的变化,碱性磷酸酶(ALP)染色法检测ALP的分泌,RT-PCT法和Western Blot 法检测细胞内促终末期分化因子基因mRNA和蛋白的表达.
结果: 人骨关节炎软骨细胞具有终末期成熟分化的特征,与对照组相比,PTH明显降低ALP染色的强度,显着降低细胞内促终末期分化基因mRNA和蛋白的表达.
结论: PTH具有抑制人骨关节炎软骨细胞终末期分化的作用. |

Investigation of the anti-terminal maturation effects of parathyroid hormone on human osteoarthritis chondrocytes |

ABSTRACT
Objective: To investigate the inhibition in parathyroid hormone(PTH) to terminal differentiation of human osteoarthritis chondrocytes.
Methods: Human osteoarthritis chondrocytes were isolated and cultured. The cells were divided into PTH group and control group,and the cells of two groups were treated respectively with PTH and saline every day. The morphology of cells was observed by microscope. Alkaline phosphatase(ALP) staining was performed to detect the ALP protein. The hypertrophic marker genes mRNA and protein expressions were detected by real-time RT-PCR and Westernblot.
Results: Human osteoarthritis chondrocytes had characteristic of terminal differentiation. Compared with control group,PTH group can obviously degrade ALP staining density,and decrease marker gene mRNA and proteinic expression of promoting terminal differentiation.
Conclusion: PTH has a role of inhibition of terminal differentiation of human osteoarthritis chondrocytes. |
