目的:研究兔成骨细胞在生物活性玻璃与TGF-β复合生物支架中的生长情况,探讨优化组织工程中生物材料支架的实验方法。
方法:体外培养的兔成骨细胞与BG/TGF-β构建的三维立体材料支架体外复合培养,分别于2、4、6、8d通过显微镜观察,上清液ALP测定,MTT细胞计数法及流式细胞仪检测,评估BG/TGF-β构建的三维立体材料支架与兔成骨细胞的生物相容性。
结果:兔成骨细胞在BG/TGF-β构建的三维立体材料支架的表面及空隙内生长状态良好,随着培养时间的延长,各组细胞数量都明显增加,各时间点BG/TGF-β构建的三维立体材料支架上MTT法测定细胞数实验组与对照组比较,两组对照有显著性差异(P<0.05),上清液ALP测定值实验组与对照组进行统计处理,两组对照有显著性差异(P<0.05)。细胞凋亡率(Ap)实验组与对照组进行统计处理,两组对照无显著性差异(P>0.05)。
结论:BG/TGF-β复合支架不但具有良好生物相容性,而且能促进成骨细胞的增殖与分化,为骨组织工程支架材料的改良提供了依据。 |

Experimental research of the growth of rabbit osteoblast on the composite BG/TGF-β material scaffolds |

ABSTRACT
Objective: To invest the growth of rabbit osteoblast on the composite BG/TGF-β material scaffolds and explore the expremental methods of optimized material scaffolds in bone tissue engineering.
Methods: Osteoblasts were cultured with bioactive glass ceramics together in vitro,the biological properties affected with BG/TGF-β were observed and measured by light microscope,check of alkaline phosphatase in supernatant liquid and the cell proliferation was detected by MTT assay,flow cytometer at 2,4,6,8 d.
Results: The results showed the cells on BG/TGF-β were good adhension and activity.The MTT assay showed the number of cells in the group were significantly increased with the culture time prolonged,the number of cells on BG/TGF-β was higher than the control group(P<0.05).Check of alkaline phosphatase in supernatant liquid showed the number of the ALP in the BG/TGF-β group was higher than the control group(P<0.05).There was not a significant difference about apoptosis result.The BG/TGF-β group was close to the control group(P>0.05).
Conclusion: The porous composite material scaffold of BG/TGF-β not only has good biocompatibility,but also can progressed osteoblasts growth and dividing. |
