目的:观察维生素D受体(VDR)、核心结合因子α1(Cbfα1)mRNA在大鼠成骨细胞(OB)中的表达,研究固本壮骨胶囊、金匮肾气丸、补肾益精方、知柏地黄丸四种补肾中药对大鼠成骨细胞VDR、Cbfα1mRNA表达的影响,探讨补肾中药促进骨形成的新机制。
方法:6月龄大鼠成骨细胞被分离,采用组织块翻转方法培养。通过倒置显微镜观察细胞形态,矿化结节染色对成骨细胞加以鉴定。采用RT-PCR方法检测成骨细胞VDR、Cbfα1mRNA表达。
结果:RT-PCR表明,在大鼠成骨细胞中,固本壮骨胶囊、金匮肾气丸、西药萌格旺上调VDR、Cbfα1mRNA表达;知柏地黄丸下调VDR、Cbfα1mRNA表达、补肾益精方对于VDR、Cbfα1mRNA表达的影响存在着差异。
结论:固本壮骨胶囊和金匮肾气丸上调成骨细胞VDR、Cbfα1mRNA表达,可促进骨形成;知柏地黄丸下调VDR、Cbfα1mRNA表达,有抑制或是降低骨形成的作用。 |

ABSTRACT
Objective:To investigate the effects of reinforcing Kidney Chinese herbal medicine such as Guben-Zhuanggu(固本壮骨),Jingui-Shenqi(金匮肾气),Bushen-Yijing(补肾益精),Zhibai-Dihuang(知柏地黄) on expression of VDR,Cbfα1mRNA and explore the underlying mechanisms.
Methods:Osteoblast of 6-month-age rats was isolated and cultured by the tissue-fragment-migrating-growth method.Biological characteristic was observed by phase-contrast microscope,mineralizing nodule staining technique and electromicroscope.The expression of VDR,Cbfα1mRNA was detected by RT-PCR.
Results:RT-PCR showed that Guben-Zhuanggu capsule,Jingui-Shenqi pill up-regulated the expression which was the same as Bone-one and Zhibai-Dihuang pill down-regulated the expression of VDR,Cbfα1mRNA in rat osteoblast,but the expression was different in Bushen-Yijing decoction.
Conclusion:Guben-Zhuanggu capsule and Jingui-Shenqi pill promoted bone formation by up-regulating the expression of VDR,Cbfα1mRNA but Zhibai-Dihuang pill inhibited the bone formation by down-regulating the expression. |
