目的:观察原发性膝骨关节炎患者滑膜细胞的体外生长特性,探讨骨性关节炎的发病机制。
方法:组织块培养法分离早期与晚期(依据Kellgren和Lawrence的放射学诊断标准划分)OA患者滑膜细胞进行培养,用光镜、电镜、组织化学
方法:等观察鉴定该细胞类型。
结果:患者滑膜细胞符合巨噬样滑膜细胞(A型细胞)及成纤维细胞样滑膜细胞(B型细胞)的特征,早期与晚期滑膜细胞在生长周期及细胞超微结构等方面存在差异。晚期较早期滑膜细胞生长周期长、生长极向性差、细胞内溶酶体丰富。
结论:组织块培养法可以有效分离人类关节滑膜细胞,建立原发性膝骨关节炎患者滑膜细胞系。 |

Biologic characteristics of human synoviocytes of gonarthritis in vitro |
|
ABSTRACT
Objective:To investigate the biological characteristics of human synoviocytes of primary gonarthritis in vitro.
Methods:Human synoviocytes were isolated from synovial membrane(earlier stage and later stage of OA) by tissue culture isolation,which were identified by light microscope,electro microscope,histochemical method.
Results:The isolated cells were in acordance with the characteristics of macrophage-like synoviocytes(type-A synoviocytes) and fibroblast-like synoviocytes (type-B synoviocytes).Differences showed in the growth cycles,differentiations and ultra microstructures between the two stages.
Conclusion:Tissue culture isolation is an effective isolation of human synoviocytes to establish the human synoviocyte line of primary gonarthritis. |
