中文摘要:
目的:观察α-平滑肌肌动蛋白(alphasmoothmuscleactin,α-SMA)在臀肌挛缩症的表达、分布。
方法:利用免疫组化染色辅以计算机图像定量分析技术,对26例臀肌挛缩症及12例正常臀肌标本α-平滑肌肌动蛋白的表达进行检测。
结果:α-SMA在对照组中未见明显表达。12例臀肌挛缩症组织中α-SMA染色阳性,14例臀肌挛缩组织中染色阴性,病变组与对照组比较差异有显著性(P<0.001);7岁以上组臀肌挛缩症组织α-SMA表达明显减弱,与7岁以下组臀肌挛缩症组织(包括7岁)比较差异有显著性(t=2.364,P=0.028)。
结论:①肌纤维母细胞是臀肌挛缩症发病的关键病理因素;②臀肌挛缩症中肌纤维母细胞的表达强度与病程有关,7岁以上组病程较长,表达明显减弱,这给临床治疗提供了新思路。 |

| Expression and significance of a alpha-smooth muscular actin in the gluteal muscle contracture |

ABSTRACT
Objective:To observe the expression and distribution of alpha-smooth muscular actin(α-SMA)in gluteal muscle contracture and discuss its clinical significance and relationship.
Methods:26 patients suffered from gluteal muscle contracture were collected from Oct.1994 to Oct.2002.The control specimens were collected from normal gluteal muscle and fascia.Expression and localization of alpha-smooth muscular actin in different groups were assessed after incubation of paraffin section with a mouse monoclonal antibody.The expression level of α-SMA was detected by computer graph analysis system.
Results:There was no significant expression of α-SMA in control group except for the positive reaction in the smooth muscular cells of the small vascular walls.12 specimens of gluteal muscle contracture were positive for α-SMA actin,while 14 specimens of gluteal muscle contracture were negative.α-SMA was expressed in the diseased group,with a striking difference from control group(P<0.001);In the gluteal muscle contracture group,the expression of α-SMA of those aged over 7 weakened obviously,whose difference from those aged less than 7 was of striking difference(t=2.364,P=0.028).
Conclusion:①Myofibroblast appeared to be one of the most important factors in the pathogenesis of gluteal muscle contracture.②The expression intensity of myofibroblast in gluteal muscle contracture was related to history of disease.In the gluteal muscle contracture group,The expression of α-SMA of those aged over 7 weakened obviously,which gives fresh thought to clinical diagnosis and cure of these diseases. |
