中文摘要:
目的:研究红曲对去卵巢大鼠BMP2表达及成骨细胞增殖的影响。
方法:取12周龄的雌性Wistar大鼠40只,其中30只打开腹腔去除双侧卵巢,逐层缝合,另10只予以单纯打开腹腔不切除卵巢。将大鼠分为A组(模型组)、B组(治疗组)、C组(阳性对照组)、D组(假手术组),饲养12周后,分别予以生理盐水、红曲水提液、αD3水溶液及生理盐水灌胃。灌胃12周后,活杀各组动物,取右胫骨上端做病理切片,通过HE染色对成骨细胞等进行观察比较、BMP2免疫组化染色观察。
结果:经过12周的红曲水提液灌胃治疗,成骨细胞数量明显增加,而且通过免疫组化染色发现有BMP2的棕色深染,阳性细胞比例明显高于模型组。
结论:红曲可促进成骨细胞增殖、分化,推测其治疗骨质疏松症的机制可能是通过提高BMP2的表达来实现的。 |

| Effects of Hongqu on BMP-2 expression and osteoblastic proliferation in ovariectomized rats |

ABSTRACT
Objective:To study the effects of Hongqu on BMP-2 expression and osteoblastic proliferation in ovariectomized (OVX) rats.
Methods:Laparotomy were done in 40 female Wistar rats with 12 weeks age,among them,ovaries of 30 rats were cut off,the others did not deal anything.They were divided into four groups:Model,therapy,positive control and sham operation group (group A,B,C,D).Saline,Hongqu extractant,α-D3 aqueous solution,saline were given respectively during 12 weeks after operation.All rats were killed at 12 weeks after administration.The proximal end of the right tibia were cut for pathological section,and osteoblasts were observed with HE staining and BMP-2 expression in bone tissue by immunohistochemistry staining.
Results::Compared with group A,the expression of BMP-2 and the number of osteoblasts were significantly increased in group B.
Conclusion:Hongqu obviously increased the number of osteoblasts in OVX rats.The Results showed that high expression of BMP-2 may be an important therapeutic mechanisms in osteoporosis. |
