目的:比较检测风寒湿痹证型颈椎病动物模型中颈椎间盘Fas、bcl-2蛋白的表达;检测葛根汤、桂枝汤对风寒湿型颈椎病家兔颈椎间盘组织中Fas、bcl-2蛋白的调节作用。
方法:8月龄雄性新西兰白兔24只,随机分为正常对照组、风寒湿刺激组、葛根汤治疗组、桂枝汤治疗组。风寒湿刺激组温度6℃,湿度95%,风力6级,每日连续刺激4h,间断重复刺激128h;葛根汤、桂枝汤治疗组在风寒湿刺激组造模基础上服用。采用免疫组织化学ABC法检测椎间盘石蜡切片中Fas、bcl-2蛋白的表达情况。
结果:风寒湿刺激组同正常对照组比较,Fas表达均上调(P<0.01),葛根汤降低Fas表达,与风寒湿刺激组比较有显著性差异(P<0.01)。风寒湿刺激组同正常对照组比较,bcl-2表达均下降(P<0.05~0.01),葛根汤上调bcl-2表达,但与风寒湿刺激组比较没有显著性差异(P>0.05),桂枝汤对bcl-2的调节作用不明显。
结论:风寒湿刺激组同正常组对照比较,Fas表达均上调,bcl-2表达均下降,葛根汤降低Fas表达,上调bcl-2表达,发挥延缓椎间盘退变的作用,桂枝汤调节上述细胞因子的作用低于葛根汤。 |

Expression of the Fas receptor,bcl-2 in the cervical intervertebral disc of the rabbit modified by the Guizhitang and Gegentang |

ABSTRACT
Objective:To compare the expression of the Fas receptor,bcl-2 in the degenerative intervertebral disc of the rabbits cervical spondylosis model induced by Fenghanshi.To study the effects of the Guizhitang,Gegentang on the expression of the Fas receptor,bcl-2.
Methods:24 male rabbits were divided randomly into normal group,Fenghanshi group,Gegentang group and Guizhitang group.Fenghanshi model was created in 6 ℃ temperature,95% humidity,and 6 grade wind with 4 hours stimulation at each day for the total of 128 hours.Fenghanshi group was treated by Gegentang and Guizhitang.The expression of the Fas receptor,bcl-2 in the degenerative disc was detected with the immunohistochemistry ABC.
Results:The expression of the Fas receptor increase in the Fenghanshi model compare with the normal (P<0.01).Gegentang could regulate down the expression of Fas receptor in the fenghanshi model (P<0.01).On the other hand,the expression of the bcl-2 decrease in the Fenghanshi model compared with the normal (P<0.05~0.01).Gegentang could regulate up the expression of Fas receptor in the Fenghanshi model (P>0.01)
Conclusion: The expression of the Fas receptor can be regulated up and the bcl-2 regulated down in the Fenghanshi model compared with the normal.Gegentang can regulate down the expression of the Fas receptor and regulate up the bcl-2. |
