目的:设计制作静脉回流受阻骨坏死动物模型,探讨Legg-Perthes病发病机理。
方法:幼犬20只,应用医用TH胶股骨颈内注射法制作LeggPerthes病动物模型,于注药后4周、8周、12周分别行X线及CT检查,并分批处死动物观察组织学改变。
结果:注药后其影像学改变最早发生于术后8~12周,在早期即可出现类似Legg-Perthes病的表现。组织学改变表现为骨小梁变稀疏,骨细胞脂肪变性,骺板变薄,骺板内软骨细胞排列不规则。
结论:应用医用TH胶股骨颈内注射法制作Legg-Perthes病动物模型,探讨静脉回流受阻因素在Legg-Perthes病发病中的作用机理,方法简单,效果可靠。 |

Experimental study on the animal model of Legg-Perthes disease |

ABSTRACT
Objective:To establish an animal model of osteonecrosis induced by obstruction of venous reflux and investigate the pathogenesis of Legg Perthes disease
Methods:Medical TH adhesive was injected into the femoral neck of 20 healthy juvenile dogs then a model of Legg Perthes disease was set up.X ray and CT study were performed on the 4,8,12 weeks after drug administration respectively.Then,the dogs were sacrificed and the histological changes were studied.
Results:The earliest changes in image after drug administration took place in 8-12 weeks postoperatively.CT change presented more significantly and earlier than that seen on the X ray film,which is similar to the pattern of Legg Perthes disease at the earlier stage.Sparsed bone trabeculae,fat degenerated osteocytes,thinned epiphyseal plate and irregular arranged chondrocytes of epiphyseal plate can be observed in histological study.
Conclusion:This a simple and reliable method by the injection of medical TH adhesive to set up a Legg Perthes disease model,which facilitating to investigate the mechanism of the obstruction of venous reflux in the process of Legg Perthes disease. |
