目的:为进一步探讨健骨颗粒治疗原发性骨质疏松症的作用机理,拟从垂体甲状腺轴角度进行研究。

方法:建立6月龄SD雌性大鼠卵巢摘除骨质疏松模型,模型成立后随机分为Ⅰ、Ⅱ、Ⅲ三组,分别喂服生理盐水、骨松宝和健骨颗粒。喂药12周后处死取材,取血清检测TSH、FT3、FT4、CT等浓度,取垂体、甲状腺作TSH细胞、C细胞及Tg等免疫细胞化学染色,并形态计量。

结果:喂服中药组的血清TSH、FT3、FT4、CT浓度均明显上升,与生理盐水组相比有显著统计学意义。Ⅱ、Ⅲ两组TSH细胞和C细胞的细胞数均高于Ⅰ组。Ⅲ组Tg阳性反应物也明显较Ⅰ组多,颜色深。

结论:健骨颗粒能有效扭转卵巢摘除后垂体甲状腺轴功能减退的趋势,改善甲状腺的整体功能,从而增加骨形成抑制骨吸收,达到治疗骨质疏松症的目的。

Effect of Jiangu granula on the Pituitary-Thyroid axis of ovariectomized rats
ABSTRACT  

Objective:To investigate the effect of Jiangu granula on the treatment of primary osteoporosis(OP) from the pituitary thyroid axis

Methods:OP model animals by ovary excised were randomly divided into 3 group:normal saline group Ⅰ,Gusongbao treating group Ⅱ,Jiangu granula treating group Ⅲ.The animals were sacrificed after 12 weeks to collect experimental samples.The levels of TSH,FT3,FT4 & CT in serum were measured.TSH cell in pituitary,C cell and immune globulin in thyroid gland were detected by immunocytochemistry and morphometric observations.

Results:The levels of TSH,FT3,FT4 and CT in treated groups were increasing comparing with normal saline group.The amount of TSH cell and C cells in group Ⅱ and group Ⅲ groups were more than group Ⅰ.The Tg positive reaction in group Ⅲ were also more and the colour was deeper.

Conclusion:Jiangu granula can cure primary osteoporosis,by means of turning round the functional decrease of pituitary thyroid axis after the ovary excised,and improve effectively the whole function of thyroid gland,and then raising the bone formation and reducing the bone resorption.
