髓复康促进大鼠脊髓修复再生的超微结构研究 |

目的:观察脊髓损伤后,脊髓神经毡等超微结构的变化和中药髓复康对于这种变化的影响;另外,希图从脊髓损伤区毛细血管内皮细胞的超微结构方面,探查髓复康对大鼠损伤脊髓的保护作用、促修复再生作用及其作用机理。
方法:制备45只下胸髓右侧半横断损伤的Wistar大鼠模型,分为髓复康、补阳还五汤、甲基强地松龙和空白对照4组,另设正常对照组。用透射电镜和图像分析技术观察和分析髓复康对脊髓损伤区神经元和毛细血管内皮细胞超微结构的影响,探讨其与受损神经组织修复再生的关系。
结果:①在空白对照组大鼠,脊髓损伤区神经元胞体、轴突和树突野的超微结构发生明显的变性。及时服用髓复康可以保护脊髓损伤区的神经元,并且促进受损神经元胞体、神经纤维和树突野的修复再生:②空白对照组大鼠脊髓损伤区毛细血管内皮细胞的饮液小泡数量明显减少,而服用髓复康的大鼠,饮液小泡的数量增加;③髓复康组大鼠的脊髓星形胶质细胞和毛细血管周细胞的反应比较轻微。
结论:脊髓损伤后及时服用髓复康可以:有效地降低继发性脊髓损伤,减小脊髓损伤波及的范围;增强脊髓损伤区毛细血管内皮细胞的转运功能,迅速消除损伤区的水肿,对伤区的神经元起保护作用;抑制神经胶质和周细胞的反应性增生,创造有利于神经元再生的微环境。 |

Ultrastructure study of spinal cord restoration and regeneration promoted with Chinese herb(suifukang)in rats |

ABSTRACT
Objective:Ultrastructural alterations of spinal neuropile after spinal cord injury (SCI) and the effect of Suifukang(SFK),a Chinese herb decoction,on the alterations were observed The ultrastructure alterations of spinal capillary induced by SCI,and the effect of the SFK on the alterations were observed in order to investigate protective effect of the SFK of spinal cord trauma and the possible mechanism of promoting restoration and regeneration of injured spinal tissue from the point of modulation upon neuron and capillary endothelium ultrastructure
Methods:Models of right hemitransection at the lower thoracic spinal cord were made in 45 Wistar rats and the animals were divided into 5 groups: ①SFKG After the spinal hemisection,12 rats were administrated with SFK from one day to a month;②BYTG:After the spinal hemisection,12 rats were administrated with Buyanghuanwutang (as control Chinese herb)from one day to a month;③MPSG:After the spinal hemisection,9 rats were treated with methylprednisolone from one day to a month;④BCG:After the spinal hemisection,12 rats were administrated with NS from one day to a month; in addition,there was ⑤NCG:3 normal rats uninjured the spinal cord For groups 1~4,12 rats were subdivided into 4 subgroups:the rats were treated for 1,3,15,and 30 days respectively,but there was no 30 day subgroup in the BCG specimens The injured areas were procured from the spinal cords,and prepared into ultrathin sections,and observed with TEM These electromicrophotographs were analyzed with regeneration of spinal neurons
Results:There was obvious ultrastructural degeneration of spinal neuron,axon and dendrite profile in BCG It has been observed that the protection for spinal neurons and regeneration of the perikaryon,axon and dendrite profile was distinct in rats treated with SFK After spinal cord injury,amount of pinocytosis vesicles in capillaryendothelial cells reduced distinctly in BCG,and the amount was increased in SFKG Degree of reaction of the glia and pericytes to spinal injury was much less in rats SFKG
Conclusion: After spinal cord injury and administration with SFK,it has been observed that:①The secondary spinal cord damage was distinctly reduced and spinal injury area was smaller ②the transport function of endothelium was reinforced and traumatic edema of the cord subsided rapidly ③It is possible that these alterations possess protective effects on spinal neurons and the recactive proliferation of glia and pericytes was inhibited The microenvironment that adapts regeneration of spinal neurons induced as a result of enhancement of regeneration of the nervous tissue |
