椎间盘纤维环中糖胺多糖的实验研究

目的:研究正常和退变腰椎间盘纤维环中糖胺多糖(GAG)含量及其主要成份百分组成,探讨椎间盘退变的发生机理。

方法:对34个正常纤维环和35个椎间盘突出症纤维环标本中GAG含量进行测定,并用醋酸纤维素膜电泳和光密度扫描的方法对GAG主要成份百分组成进行测定。

结果:正常纤维环GAG含量为4.17±0.94g/100g,退变纤维环GAG含量为3.57±0.98g/10.0g,低于正常组(P<0.05);退变组GAG中硫酸软骨素(CS)百分值低于正常组(P<0.05);而其透明质酸(HA)和硫酸角质(KS)高于正常组(P<0.05

结论:椎间盘纤维环中GAG含量下降,是引起纤维环胶原纤维稳定性降低,影响纤维环板层滑动,导致椎间盘退变、突出症发生的重要原因之一。退变纤维环GAG含量下降由其主要成分CS含量下降所致。

An Experimental Study of the Glycosaminoglycan in the Annuli Fibrosus of the Normal and Degenerated Intervertebral Disci
ABSTRACT  

Objective:To study the glycosaminoglycan(GAG)content and its percentage composition in annuli fibrosus of normal and degenerated intervertebral disci,and to discuss the pathogenesis of degeneration of intervertebral disc

Methods:The GAG content and its percentage composition were determined in 34 specimens of normal annulus fibrosus(from autopsy specimens of patients of cerebral injury and in 35 degenerated specimens of annulus fibrosus Celluloseacetate membrane electrophoresis and optical density scanning were used in this experiment

Results:The GAG content in normal group was higher than those in degenerated group(P<0.05);The percentage composition of chondroitin sulfate(CS) in the degenerated group was lower than that in the normal group(P<0.05);The percentage composition of hyaluronic acid(HA)and keratin sulfate(KS)was higher than that in the normal group(P<0.05)

Conclusion:The decrease of GAG content in annuli fibrosus plays an important role in the decrease of stability of collagenous fibers1It also affects sliding of annuli fibrosus lamella and leads to the degeneration and herniation of intervertebral disc1 The decrease of GAG content in the degenerated annuli fibrosus is mainly caused by the decrease of CS.
